Police attended the scene after a van was seen entering the underground car park of a hotel building on Duncans Road at the Air Wing.
The air wing had been assisting officers on the ground after the driver of the van sped away from police in Werribee at around 9.30pm.
Police believe the deceased man was the driver of the van.
Police said they were investigating a possible connection between the van and a grand theft carjacking in the nearby suburb of Hoppers Crossing on October 22.
Homicide detectives will investigate the incident under the supervision of the Professional Standards Command, as is usual when a person dies in police presence.
